演題:Structure and Dynamics of Rad51 filament during homology search in living cells
要旨:In eukaryotes, the recombinase Rad51 plays a central role in Homologous recombination by forming nucleoprotein filaments (NPF) on single-stranded DNA (ssDNA) to scan the genome and invade a homologous sequence which will be used as a template for DNA repair synthesis. Although Rad51 has been extensively studied by in vitro, molecular, and genetic approaches, its dynamics in vivo could not be assessed until recently due to the lack of functional tags. Our team has developed the first fully functional version of Rad51 in budding yeast revealing the formation of dynamic, micrometer-long filaments following the induction of a unique DSB (Liu et al., 2023). Once formed, Rad51 filaments are surprisingly dynamic, undergoing cycles of compaction and extension events. Physical modeling demonstrate that such dynamics represent a robust and efficient mechanism of homology search. I will discuss our recent results on the structure, organization and regulation of Rad51 filaments in living cells.
